Rapamycin (Sirolimus)

Rapamycin is a promising therapy for promoting healthspan by modulating the mTOR complex I pathway, which plays a key role in cellular aging. When taken in small doses over time, Rapamycin may aid in cellular function, slowing the accumulation of senescent cells and promoting longevity.

12 capsules/bottle. $83/mo ($249 for a 3-month supply).

Rapamycin is taken orally in capsule form. For healthspan purposes, Rapamycin is taken once weekly, rather than daily, to minimize potential side effects while still modulating the mTOR pathway effectively.

Rapamycin is generally well-tolerated when taken in lower, intermittent doses for longevity purposes. However, potential side effects can vary based on dosage, frequency, and individual response. Common side effects may include:

  • Mild digestive issues – Nausea, diarrhea, or stomach discomfort
  • Mouth ulcers or sores – A common but often temporary side effect
  • Fatigue or weakness – Some individuals may experience reduced energy levels
  • Increased risk of infections – As Rapamycin modulates immune function, higher doses may slightly increase susceptibility to infections. We require follow-up CBC lab testing to monitor WBCs.
  • Changes in lipid levels – May cause mild increases in cholesterol or triglycerides
  • Delayed wound healing – Due to its effects on cellular growth pathways

Most side effects are dose-dependent and tend to be minimal when Rapamycin is taken at low, intermittent doses for healthspan support. Working with our healthcare providers can help optimize dosing and minimize potential risks.

Yes, lab monitoring is required to ensure safety and optimize therapy. We will track white blood cell (WBC) counts through a Complete Blood Count (CBC) and Comprehensive Metabolic Panel (CMP) to assess overall health and immune function.

  • A repeat CBC will be conducted 90 days after starting therapy to monitor any changes.
  • Ongoing lab monitoring will then be adjusted to every 6 to 12 months, as determined by your healthcare provider.

Regular testing helps ensure that Rapamycin remains safe and effective for long-term use.

We prescribe Sirolimus because it is more bioavailable than the compounded version of Rapamycin.

Generally, 5 mg capsules taken once a week orally. New patients will be titrated up by clinician guidance. Providers may also cycle patients off for a short period of time.

Possible side effects include sore or inflamed mouth, weakened immune system, anemia or fatigue, constipation or diarrhea, elevated lipid levels, high blood pressure, swollen hands or feet. Please contact us on the onset of any symptom you think may be related to treatment.

Rapamycin directly stimulates the cellular cleaning process known as autophagy, effectively clearing toxic debris and promoting cellular rejuvenation.

Cellular senescence is a process that results from a variety of stresses and leads to a state of irreversible growth arrest. Senescent cells accumulate during aging and have been implicated in promoting a variety of age-related diseases.

The mTOR pathway, found in all cells, determines when a cell should grow and divide. However, with time, it can become too active, resulting in unchecked cell growth and compromised tissue health. Rapamycin is a direct inhibitor of the mTOR pathway. Low doses of rapamycin appear to reset mTOR activity to a more youthful state—active when needed and dormant when not required.

At Joi+ Blokes, we take a holistic, preventative approach to care, which means many advanced treatments and lab tests are not typically covered by insurance. However, we do offer insurance-pay options for certain prescriptions:

  • Women’s health medications like estrogen and progesterone can be sent to your local pharmacy, where you may use your insurance benefits.
  • Thyroid medications will be sent to your local pharmacy, allowing you to utilize your insurance coverage.
  • FDA-approved GLP-1/GIP medications for weight loss and type 2 diabetes, including Ozempic, Wegovy, Zepbound, and Mounjaro, may also be eligible for coverage.

While we assist with prior authorizations and prescription submissions, we do not bill insurance directly. We recommend checking with your insurance provider for specific coverage details.

Yes! You can use your HSA or FSA funds to cover eligible purchases. We are happy to provide a detailed receipt and a Letter of Medical Necessity (LMN) from your provider. You can then submit these documents to your HSA/FSA account for potential reimbursement. Since coverage varies by plan, we recommend checking with your provider to confirm eligibility.
