Clomiphene stimulates the body’s natural testosterone production by acting on the hypothalamic-pituitary-gonadal (HPG) axis. It functions as a selective estrogen receptor modulator (SERM), blocking estrogen receptors in the hypothalamus. This prevents estrogen from signaling the brain to reduce testosterone production.

 

By blocking estrogen’s negative feedback, clomiphene triggers the release of gonadotropin-releasing hormone (GnRH) from the hypothalamus, which then stimulates the pituitary gland to increase luteinizing hormone (LH) and follicle-stimulating hormone (FSH) production.

  • LH signals the testes to produce more testosterone
  • FSH supports sperm production, maintaining fertility
